چکیده

Nav1.3 TTX-sensitive, voltagegated sodium channel Embryonic DRG and TG neurons, spinal cord, and thalamus; upregulated in DRG and dorsal horn neurons following nerve injury Loss of function attenuates pain-related behavior Na+ channel blockers including local anesthetics, anticonvulsants, antiarrhythmic, and TCAs relieve pain associated with neuropathy, diabetes, osteoarthritis, and surgery. Mutations in the Nav1.7 gene (scn9a) cause erythromelalgia and paroxysmal extreme pain disorder; loss of function of Nav1.7 results in pain insensitivity. Nav1.7 TTX-sensitive, voltagegated sodium channel Most adult DRG and TG neurons; upregulated in small DRG and TG neurons postinflammation Loss of function reduces mechanical and thermal hypersensitivity; the Nav1.7/Nav1.8 blocker CDA54 reduces neuropathic pain in nerve injury models

Simulation study of the transport properties of ions through ion channels serving as primary components of a nanobiosensor

Ion channels are naturally occurring pores through the proteins that regulate the passage of ions and thus maintain the concentration of ions inside and outside the cell. The ion channels control many physiological functions and they can show selectivity for a specific ion. Ion channels are mostly observed in nerve cells and muscle cells. The influx of ions into cells can be regulated by a gate...

Ion channels and osteoarthritic pain: potential for novel analgesics.

Osteoarthritis (OA) is a debilitating chronic condition widely prevalent in ageing populations. Because the pathology of the disease includes cartilage erosion and joint remodelling, OA patients experience a great deal of pain. Despite numerous studies, details of OA are frequently inseparable from other types of chronic pain, and its causes are unknown. In most circumstances in OA, the cartila...

Acid-Sensing Ion Channels and Pain

Pathophysiological conditions such as inflammation, ischemia, infection and tissue injury can all evoke pain, and each is accompanied by local acidosis. Acid sensing ion channels (ASICs) are proton-gated cation channels expressed in both central and peripheral nervous systems. Increasing evidence suggests that ASICs represent essential sensors for tissue acidosis-related pain. This review provi...

Pain channelopathies.

Pain remains a major clinical challenge, severely afflicting around 6% of the population at any one time. Channelopathies that underlie monogenic human pain syndromes are of great clinical relevance, as cell surface ion channels are tractable drug targets. The recent discovery that loss-of-function mutations in the sodium channel Nav1.7 underlie a recessive pain-free state in otherwise normal p...
